In the present study, we investigated whether the lack of bicarbonate-dependent reabsorption of tubular proteins, the well-known cause of chronic renal insufficiency in the X-linked hereditary renal tubular disorder of the anuric rat, could affect the expression of IL-2R on the surface of activated T cells. The expression and distribution of IL-2R on the surface of activated T cells was studied by immunofluorescence, and the expression and affinities of the receptors were analyzed by Scatchard plots. To activate T cells, anti-CD3 and anti-CD28 antibodies or phorbol 12-myristate 13-acetate (PMA) were used. Kinetics of IL-2R expression and occupancy were studied after PMA activation. The expression of IL-2R, as measured by fluorescence intensity, was reduced on activated T cells from anuric rats as compared to T cells from controls. In vitro activation of T cells with anti-CD3 and anti-CD28 antibodies increased IL-2R expression on T cells from both normal and anuric rats. The inhibitory effect of anuria on IL-2R expression was reversed by bicarbonate reabsorption in vitro. IL-2R expression increased in the presence of PMA, and this effect was detectable after as little as 5 min of activation with PMA. Bicarbonate did not alter this effect. The increase in IL-2R expression after PMA activation was accompanied by the appearance of IL-2 binding sites. A polyclonal murine anti-rat monoclonal anti-IL-2R antibody, used in Scatchard plots, detected both high- and low-affinity binding sites. A calcium-binding protein (CAP) has been isolated from skeletal muscle of fishes belonging to two different families (alvine and thunnus). The protein has been purified by ion-exchange chromatography on Dowex 50-WX8 and gel filtration on Sepharose 6B. The molecular weight of CAP has been determined by SDS polyacrylamide gel electrophoresis to be 24,000 daltons. Purity of the preparations was checked by gel electrophoresis and by monitoring the fluorescence intensity of the tryptophan residues which is directly correlated with the change in protein conformation in response to calcium binding. Electron microscopy showed that the protein was aggregated in the presence of calcium. CAP isolated from alvine and thunnus muscle are extensively cross-reactive with a commercially available antiserum directed against rat skeletal muscle CAP.
